It seems like would lose 200 points. Do the math yourself. The measure that the SP ends on is larger than the one that the SP starts on.

It stands for Full Combo, it's when you get 100% in a song without losing your streak i.e. without overstrumming.

NickOfTime146's video of this song is currently first place across all platforms, so I can assume the path he uses is optimal (or at least close to it). Here's his path in text (credit goes to him, of course):
